The Miniprep Song

Minipreps...
Have an awful lot of spinning steps
But it seems there's not a better way
Of harvesting the DNA.

Resuspending cells is quite a chore
My piptetting thumb is getting sore

Qia kits are great, 'cause I hate the phenol way
Still, next time you clone, clone by phone, it saves a day (hey hey hey)

When you've finally gone through all the steps
It's time to do the maxipreps!

I love doing minipreps. They keep you busy, they always work, you generally get useful information back. The qiagen kit is expensive, and overkill. I follow:

Very reliable. If you need it cleaned up more (sequencing?) you can add a touch of RNAse and put it over a spin column, but for restriction digests, they're fine.
